From Route 22:
- Take Route 378 South for 3.6 miles. After you pass the third exit, move over to the left lane.
- Turn left at the first stop light onto Third Street.
- Go through three stop lights and turn left onto Webster Street.
- Go straight through the stop sign, and OraSure Technologies corporate headquarters will be the red brick building on your right.

From Route 78 East or West:
- Exit I-78 at the Hellertown/Bethlehem exit 67 (formerly Exit 21).
- Turn right at the end of the exit ramp onto Route 412 North. You will go under two overpasses. Bear left at the fork staying on Route 412 North. Bear right to stay on Route 412 North until it turns into Third Street.
- Turn right at the stop light on Webster Street.
- Go straight through the stop sign, and OraSure Technologies will be the red brick building on your right.

From Points South:
- Take the Northeast Extension of the Pennsylvania Turnpike (Route 476) North to Quakertown, Exit 32.
- Turn left onto Route 663 and follow it for 3.5 miles.
- Turn left onto Route 309 North to Center Valley.
- Turn right onto Route 378 North towards Bethlehem over South Mountain to the Third Street stop light.
- Turn right onto Third Street.
- Go through three stop lights and turn left onto Webster Street.
- Go straight through the stop sign, and OraSure Technologies will be the red brick building on your right.
